Is it possible to create lines and curves in Creo Illustrate without specifying a reference. A little bit like markups in Creo View ?

It is possible but not in a direct way.
The method I use is a temp part. I put a wrench or arrow etc from my symbols library where I need to put a point, a line or something. Once you have that point or line. You can move or remove the temp part. It is only needed for the reference points to be created.
Remember that you need a reference for all 3 dimensions in order to create a new element in the 3D space.
