I have a fixed - free bar with tichness 40 [mm].
the deflection is 0,2 [mm] with F = 4.000,00 [N]
In Simulate I can investigate the value of thickness in order to not superate 0,1 [mm] of deflection?
Maybe I need a sensitivity analysis, correct?

yes that will work or you can do an optimization something like this.
This is a CAD problem, not a simulate problem. It means the dimension is dependent on something so it cannot be modified. Can you change the thickness manually to different thicknesses? Are you using a relation (equation) to drive the thickness?

can I not create "optimization study" with 3D model regenerate from Creo Modeling?

You should be able to do this. Do you have Creo 5? I can send an example solid beam model with thickness as a variable.
