I drafted a drawing for an assembly and used a iso tolerance at a particular dimension but when I upload and checked in it to wind-chill I wasn't able to see that particular dimension in iso tolerance . It's not getting updated even on model prepare regen

As that's a property of the dimension, which I'll presume is a feature dimension and thus lives in the solid, look to Windchill for the revision history of the solid.  I'm going to guess that your changes to the solid aren't there, either because 1) in checking in the drawing+solid, you had a warning about conflicts, and opted to discard changes to the solid but check in the drawing anyway (which is a bad technique unless you are quite sure of what you are doing), or 2) someone else later checked in a version of the solid that did not include your changes, and was able to overwrite your changes with theirs.
