Temperature and duty cycle in Calculation data tab

If I want to run a prediction of a system with a specific temperature and duty cycle, should I enter those values in Calculation data tab, too, as well as running a mission profile? Or should the temp and duty cycle usually be 25C and 100%, and then use the Mission profile to run different temps and duty cycles?
